I have an application of using HPLC and UPLCs to analyze pharmaceuticals. Im being asked if ultrapure water is needed for this application or if USP purified water could be used and why?
Answer
The deciding factor is the amount of dissolved CO2 and the organic contamination levels in the water.
USP Purified does not have a strict TOC level and would usually be unsuitable for HPLC as it would raise the baselines.
Lab polishing systems for HPLC use a 185 nm ultraviolet light and special scavenger resins to deal with to deal with the TOC and have scavengers for removing the CO2.
